Investigating the effect of melatonin and in L-carnitine on sperm quality parameters, inductionof DNA fragmentation and membrane lipid peroxidation during the sperm freezing process

Introduction: Sperm freezing is a procedure used to preserve sperm cells. However, freezing-thawing of spermaffects the physiological function and shape of sperm. Adding antioxidants to semen increases the quality offrozen semen. The aim of this study is to investigate the effect of antioxidants melatonin and L-carnitine onsperm parameters in healthy individuals after freezing and thawing.Methods: A number of ۴۰ volunteers went to the histopathology laboratory of Dr. Farzam. The samples weretransferred to the embryology laboratory of Qazvin University of Medical Sciences and finally they weredivided into ۴ groups, including ۱- group which only had a freezing medium. ۲- The group to which theantioxidant melatonin was added to a concentration of ۱۰ micromoles before freezing. ۲- The group to whichthe antioxidant L-carnitine was added to a concentration of ۱۰۰ micromoles before freezing. ۴- The group towhich the concentrations of both antioxidants were added before freezing Said added. All groups were frozenand thawed after two weeks. Finally, sperm parameters were evaluated and the results were analyzed byANOVA method in SPSS.Results: The present study showed that the combination of melatonin and L-carnitine in the sperm freezingenvironment can have a significant effect (P<۰.۰۵) on the parameters of the sperm except morphology. Motilityand viability: (Increase from ۱۵% to ۲۵%), DNA fragmentation (decrease from۳۲% to ۲۳%) and MDA level(decrease from ۱۰ to ۶.۵ μm/dl had significant changes.Conclusions: The results of this study show that the combination of these two antioxidants can be used insperm freezing in infertility centers.
